2.4.Lipidperoxidationconcentration
Lipidperoxidationinroottissuewasdeterminedbymeasuringmalondialdehyde(MDA),amajorthiobarbituricacidreactivespecies(TBARS)andproductoflipidperoxidation(HeathandPacker,1968).Rootsample(100mg)washomogenizedin5mlofTCA(0.1%,w/v)followedbycentrifugationat10,000×gfor10min.To1mlaliquotofsupernatantwasadded4mlof0.5%TBAin20%TCA.Themixturewasincubatedat95◦Cfor30min.Thereactionwasstoppedbycoolingovericeandcontentscentrifugedat10,000×gfor10min.Theabsorbanceofthesupernatantwasrecordedat532nm.Thevaluefornon-specificabsorbanceat600nmwassubtracted.TheamountofMDAwascalculatedusingtheextinctioncoefficientof155mM−1cm−1andexpressedasnmolg−1FW(freshweight).
